The Protein Structure Factory (PSF) features a collaborative, »conveyor belt«-like approach to protein structure determination where each step is carried out by a specialized group. See a description of the PSF and its subprojects for an outline.

The Protein Structure Factory studies the structures of human proteins. Target lists are compiled in collaboration with the group of Peer Bork at the EMBL Heidelberg. The German Resource Center provides the cDNA starting material. cDNA fragments are cloned into yeast and E. coli expression vectors. High throughput methods for cloning and characterisation have been established.

E. coli fermentation and large scale purification are carried out by Alpha Bioverfahrenstechnik.

The goal of the E. coli expression subproject is to supply the PSF with the appropriate E. coli expression clones. See »what we do« for a more detailled description of our methodology.

Activities of the E. coli expression group include:

  • construction of suitable expression vectors
  • development of protein expression assays
  • development and optimization of protein purification procedures
  • development of automation techniques for HTP cDNA cloning and HTP protein expression and —purification
  • production scale generation of cDNA expression clones and expression testing
  • implementation of a LIMS for bookkeeping experimental results as well as properties of the generated clones which is integrated in the PSF wide information storage system.
