Abstract

The term "Protein Structure Factory" stands for a common initiative of structural biologists from the Berlin area and the German Human Genome Project (DHGP). The Protein Structure Factory represents an integrative approach to the high-throughput analysis of three-dimensional protein structures.
We have set out to establish an analytical infrastructure to characterize human proteins whose cDNA sequences are available at the Resource Center of the DHGP in Berlin. Target proteins are selected on grounds of predicted novelty of the structure as well as medical or biological relevance. We combine sequence analysis with bioinformatics, automated gene expression and protein purification, determination of a biophysical fingerprint and finally the three-dimensional protein structure analysis by either NMR spectroscopy or X-ray diffraction techniques. The crystal structure analysis will use the synchrotron radiation of the storage ring BESSY in Berlin.
